FAQs

Can I use any plasmid?

Many plasmids that work in E. coli are compatible with Forager-1. However, some plasmid origins are not efficiently recognized and not every selection marker works identically. Confirmed origins include pMB1, ColE1, pUC, and RK2, and selection with carbenicillin, kanamycin, and chloramphenicol markers work well. We are always testing and optimizing these here at Forage, check the latest manual for details.

Don't I need to recover in rich media after incubating with DNA to express the selection marker?

We generally do not recommend a recovery between transformation and plating. In our proprietary media, Forager-1 is competent and metabolically active, so transformation and selection marker expression happens in a single step. Since Forager-1 doubles rapidly in rich media, replication during a prolonged recovery is likely to inflate the apparent transformation efficiency. However, for some difficult plasmids, a brief recovery (~30 minutes) may be beneficial.

Why can't I store plates in the fridge?

V-nat is known to have poor cold tolerance. We know its not ideal, but plates can be stored at room temperature for several days, and once a glycerol stock is made fresh colonies are always just 6 hours away!

Can I use Forager-1 for protein expression?
